社会活动

研究领域

1)蛋白质工程的研究

发展各种蛋白质工程手段,致力于水解酶类的分子改造,具体如下:基于理性设计,实现了玉米赤霉烯酮降解酶、PG酶、内毒素降解酶、淀粉酶等酶的活性和稳定性的提高;基于半理性设计实现了木聚糖酶、果胶酶活性、稳定性的提高,和水解产物的改变;基于酵母表面展示结合分选型流式细胞仪进行高通量筛选,实现了HRV3C蛋白酶和SUMO蛋白酶切割位点的改变,实现了医用蛋白融合标签的无痕切除。

2)精通大肠杆菌、枯草杆菌和毕赤酵母表达系统。

获得优秀的酶突变体以后,如何廉价的生产出来就需要构建一个高产的分泌表达工程菌株,因此申请人带领团队致力于大肠杆菌,枯草杆菌和毕赤酵母三个表达系统的优化,具体包括:大肠杆菌分泌表达和自裂解表达系统,能在目标蛋白大量表达之后,实现可控的菌体的自裂解释放目标蛋白;构建了枯草杆菌的基因编辑系统和双向调控系统,通过上调分子伴侣,下调蛋白酶,实现了枯草杆菌的同时双向调控,可以大幅提高异源蛋白的表达量;建立了通用的毕赤酵母高效表达策略,实现了多种异源蛋白的分泌表达和成果转化;构建了大肠杆菌,酵母的直接表面展示和间接表面展示系统,实现了基于细胞表面的级联催化和蛋白酶的高通量筛选体系。

3)生物传感

  真菌毒素,环境污染物的酶降解产物是否还有毒性常规通过细胞或者动物实验来进行,耗时耗钱。而通过微生物细胞来检测产物的毒性省时省力、微生物的培养成本也低廉,为降解产物的毒性检测提供了一个便利的检测体系。通过转录因子、启动子改造,宿主的筛选构建了水杨酸,苯甲酸及其衍生物的大肠杆菌检测体系,灵敏度比报道的提高了100-1000倍;通过核受体的改造,基于蛋白酶激活的级联放大系统,构建了雌激素等小分子化合物的酵母传感细胞,首次利用该传感系统筛选到了玉米赤霉烯酮真正的脱毒菌株。
